A Study on Effect of Amiodarone on the Pharmacokinetics of Oral Hypoglycaemic Agents in Normal Rabbits.

ABSTRACT:
The present study was aimed to find out the effect of multiple dose (7 days) treatment of amiodarone, an antiarrhythmic drug on the hypoglycaemic activity of pioglitazone and nateglinide in normal rabbits. The study was intended to determine the pharmacodynamic parameters of drug interaction between pioglitazone, nateglinide and amiodarone in normal rabbits. The experiment was conducted in four stages. Six normal rabbits were selected for each stage of experiment. These studies were conducted in the same group of rabbits after the washout period of the administered drug (Pioglitazone 45 mg/ Kg body weight, Nateglinide 370 mg/kg body weight, and Amiodarone 100mg/Kg body weight) i.e. after the complete elimination of the drug. The drugs were administered orally. The blood samples were collected by marginal ear vein at predetermined time intervals and glucose levels were estimated using GOD/POD method. The results indicated that multiple dose treatment of amiodarone an antiarrhythmic drug altered the hypoglycaemic activity of pioglitazone and nateglinide when administered alone and along with pioglitazone and nateglinide in normal rabbits. This may be due to the pharmacokinetic interaction of amiodarone with pioglitazone and nateglinide. The preliminary studies indicate the combination may be unsafe in diabetes associated with arrhythmia.
